[principles]DNA and RNA could be dyed differentcolors by the solution mixed withmethylgreen and pyronine because of thedifferent polymeric degree. DNA is dyedto be green and blue by methylgreen forits high polymeric degree, whereas RNAis dyed to be red by pyronine for its lowpolymeric degree

[Contents and ManipulationsPreparing a smear of blood cells fromthe toadPut the toad into death -→open the coelom andpericardiac muscle by the scissors → dip one drop ofblood near the end of a clean slide →press the blooddrop with the end of the other slide-→ push the twoslides in opposite direction → make the blood dropform blood membrane→air dry it
[Contents and Manipulations] 1. Preparing a smear of blood cells from the toad ⚫ Put the toad into death open the coelom and pericardiac muscle by the scissors dip one drop of blood near the end of a clean slide press the blood drop with the end of the other slide push the two slides in opposite direction make the blood drop form blood membrane air dry it
FixingPlace the blood smear into 70% ethanol for 5-10mins, then air dry it .Dyeing3.Put the slide into the solution mixed withmethylgreen and pyronine for 10-15mins,then wash it by tap water.ExaminingObserve the slide under light microscope
